Construction of an S-layer neoglycoprotein with the capsular polysaccharide of Haemophilus influenzae type b
Abstract
The ultimate goal of this project is the construction of a recombinant modell vaccine against Haemophilus influenzae type b. First, the expression of genes encoding capsular polysaccharides of Gram-negative bacteria in Gram-positive, S-layer covered Bacillaceae has to be studied. For this purpose, an E. coli - Bacilus subtilis shuttle vector plasmid is going to be constructed. The suitability of this construct for the transfer of several glycosyl transferases which participate in the biosynthetic pathway of the E. coli capsule biosynthesis is investigated. The functionality of the transferred genes is analyzed in respective E. coli mutants and in B.subtilis. Enzyme activities are assayed and gene products are isolated and characterized.
