Lipid-activated sugars as reference substances for the characterization of biosynthesis intermediates of the S-layer glycan from Geobacillus stearothermophilus NRS 2004/3a (H-148/2001)
Abstract
The goals of this project are the preparation of undecaprenylpyrophosphate-bound mono- and oligosaccharides by molecular biological methods using corresponding genes from Gram-negative, LPS producing strains, and the naturally occurring acceptor lipid in the E. coli system. Isolation and purification protocols will be developed in order to obtain well characterized reference substances, which will be needed for identification of lipid-activated indermediates participating in the biosynthesis of the S-layer glycoprotein of Geobacillus stearothermophilus NRS 2004/3a.
