Microbial and molecularbiological characterisation of recultivation layers under methane fumigation
Abstract
Landfill covers are subject to landfill gas emissions. Hence, they are characterized by a distinct vertical layering depending on the oxygen status of the soil. The project investigates the microbiological community with an emphasis on methanotrophs at different depth of the recultivation substrate under landfill gas fumigation and landfill leachate irrigation. A lysimeter experiment was conducted over the years 2002 and 2003 at the ARC Seibersdorf research. The lysimeters were filled with a mixture of biowaste compost and gravel, fumigated with artificial landfill gas consisting of 60 Vol% methane and 40 Vol% carbondioxide and irrigated with landfill leachate and water respectively. 4 treatments with 4 replicates each were implemented: A) no landfill gas + water B) no landfill gas + landfill leachate C) landfill gas + water D) landfill gas + landfill leachate Soil columns were sampled in September 2003. Molecular biological (16S rDNA – PCR) and microbiological methods (Phospholipidacid pattern, repiratory quinone) will be compared and used to identify the microbial community at different depths.
