Abstract:
IDF (International Dairy Federation) and ISO (International Organization for Standardization) have developed a standardized procedure for the evaluation of the working performance of microbiological laboratories . This procedure at present provided as a draft should enable to give some clear estimate about methodologies and laboratory personnel quality. In this study, yoghurt was used as an example to assess above mentioned criteria, whereas a culture technique using surface spread and pour-plate was applied. A statistical evaluation of the countable results was made. In the second part of the thesis the growth behaviour of six Bifidobacterium strains was tested in a agar medium containing special antibiotics. The addition of an antibiotic mixture failed to deliver reliable results. Mupirocin as an alternative did not exert any significant negative influences on the growth of six Bifidobacterium strains. Therefore it can be recommended as a suitable antibiotic for suppressing the accompanying micro flora. Furthermore, a commercially available medium was compared with its manually prepared analogue for their possible differences regarding its elective performance. No significant difference was observed, however the production of the self made medium was labour-intensive.
