- Recombinant Adeno-Associated Virus (rAAV) particles are a widely used viral vector in gene therapy clinical trials. For in vivo administration, it is crucial that the vectors are available in a highly pure and non-aggregated form. During the purification process those attributes can be measured with Dynamic Light Scattering (DLS). A new buffer can be screened for aggregation before being implemented or the purified product can be monitored at the end of the process. In this Master’s Thesis mostly empty rAAV8 and full rAAV9 vectors at a concentration of 1013 particles/ml, which corresponds to reported dosages in clinical and pre-clinical studies, were tested and compared in a variety of buffers that are or were in use during a purification process. The hydrodynamic radius (Rh) of the capsids was between 13.1 and 14.3 nm for monodisperse (= non-aggregated) samples. rAAV8 vectors were found to be less aggregated than rAAV9 vectors in 12 out of 13 buffers at room temperature (+25°C). However, rAAV9 showed a higher conformational stability in 9 out of 12 samples. The capsids of rAAV8 were prone to aggregation after five freeze and thaw cycles in all tested buffers.
