Röntgenstrukturanalyse Spektroskopie Biochemie Strukturbiologie Molekularbiologie
Forschungsschwerpunkt
Wir untersuchen die Struktur-Funktions-Beziehung menschlicher Häm-Peroxidasen mit einem Fokus auf drei Vertreter. Die Enzyme Myeloperoxidase (MPO) und eosinophile Peroxidase (EPO) erfüllen wichtige Rollen in der Abwehr von Pathogenen als Teil der angeborenen Immunantwort. In chronischen Entzündungen können ihre hoch reaktiven Oxidationsprodukte jedoch zu Schäden im menschlichen Gewebe führen. Durch eine Kombination moderner Methoden aus der Biochemie, Strukturbiologie und Molekularbiologie versuchen wir Unterschiede zwischen MPO und EPO entschlüsseln und nutzbare strukturelle Merkmale für die Entwicklung neuer selektiver Inhibitoren zu finden. Die eng verwandte Thyreoperoxidase Schilddrüsenperoxidase (TPO) bildet einen zweiten Forschungsschwerpunkt. Sie ist das das wichtigste Enzym für die Produktion von Schilddrüsenhormonen. Medizinische Relevanz hat sie jedoch auch als Autoantigen in Hashimoto Thyreoditis, bei der es zu chronischer Entzündung und Abbau der Schilddrüse kommt. Wir konzentrieren uns auf die detaillierte strukturelle und biochemische Charakterisierung dieses Enzyms um die Hintergründe der hohen Prävalenz an Autoimmunerkrankungen im Zusammenhang mit diesem Enzym zu klären.
