Genetische Charakterisierung von S-Schicht Glykoproteinen. (P12966-MOB)
Abstract
Viele prokaryontische Zellen besitzen an ihrer Zelloberfläche kristalline Zellwandschichten aus proteinhaltigen Makromolekülen, welche regelmäßige Gitter ausbilden. Untersuchungen haben gezeigt, daß S-Schichten sowohl bei Archaea (Archaeobakterien) als auch bei Bakterien (Eubakterien) glykosyliert sein können. Das Ziel dieses Projekts ist es, die genetische Charakterisierung der Biosynthese von S-Schichtglykoproteinglykanen von Gram-positiven Bakterien zu initieren. Für die Untersuchungen wurde aufgrund seiner Glykanstruktur Aneurinibacillus (früher Bacillus) thermoaerophilus ausgewählt. Die genetischen Experimente werden durch biochemische Studien an aktivierten Stoffwechselzwischenprodukten der Glykanbiosnthese von A. thermoaerophilus unterstützt. Aus den Experimenten erwarten wir Informationen über die molekulare Organisation der Biosynthese von S-Schichtglykanen in Gram-positiven Bakterien. Weiters sollen die Untersuchungen Ansätze für mögliche zukünftige genetische Manipulationen an diesen gly-kosylierten Makromolekülen liefern, um in Zukunft 'maßgeschneiderte' S-Schichtglyko-proteine für spezifische biotechnologische Anwendungen herstellen zu können.
