Engineering trichothecene resistance in plants.
Abstract
Trichothecenes are a diverse class of sesquiterpenoide secondary metabolites that are produced by a number of filamentous fungi. The trichothecene mycotoxins produced by plant pathogenic fungi are probable virulence factors in many plant diseases. In particular Fusarium diseases of economically important cereal crops like wheat and maize are a worldwide problem, since they potentially result in unacceptable levels of mycotoxin contamination of human food and animal feed. Trichothecenes act as inhibitors of eukaryotic protein synthesis. Resistant mutants of the yeast Saccharomyces cerevisiae with alterations in the target of the toxin, a ribosomal protein, are characterized. The corresponding mutations are introduced in the highly conserved genes of plants to allow testing if the introduced modified transgene is able to increase trichothecene resistance. A further goal of the project is to investigate the posssibility that a similar ribosomal resistance mechanism occurs naturally in trichothecene producing fungi.
