S-layer architectures: Extending the morphogenetic potential of S-layer protein self assembly
Abstract
Two-dimensional crystalline S(urface)-layers are the most commonly observed cell surface structures in prokaryotic organisms (bacteria and archaea). S-layers are highly porous protein meshworks with unit cell sizes in the range of 3 to 30 nm and pore sizes of 2 to 8 nm. Native or recombinant S-layer proteins have the capability to form self-assembled monolayers in suspension and at various surfaces and interfaces. The main objective of this research project is the fabrication of three-dimensional S-layer protein architectures. Based on the successful methods of coating water filled liposomes with S-layer proteins, micrometer sized hollow spherical, polyhedral and cylindrical morphologies will be created. In addition, surface engineering with polyelectrolyte layers will play an important role since it will allow us to fabricate a large variety of different surfaces using simple liquid handling procedures. It is anticipated that three-dimensional S-layer architectures will play a key role in a broad range of novel applications ranging from the development of artificial viruses, controlled drug targeting and delivery systems to novel affinity matrices.
