Detoxification mechanisms in plants: functional testing of UDP-glucosyltransferases
Abstract
The goal of this project part is to utilize the model system Brachypodium distachyon as a tool for investigation of Fusarium resistance in cereals and in particular for research on detoxification of Fusarium metabolites. The rationale is that orthologs of genes identified in this close relative of cereals should be responsible for resistance components also in complex plants such as hexaploid wheat. We plan to infect genetically highly diverse Brachypodium accessions (ecotypes) with multiple Fusarium strains (differing in origin and trichothecene chemotypes) to observe variation in fungal aggressiveness and variation in plant resistance. We will furthermore challenge the plants with the Fusarium metabolites DON and ZON.We assume that members of the very large gene family of UDP-glucosyltransferases (UGTs) play an important role in detoxification of fungal metabolites. Based on the genome sequence of Brachypodium we plan to express individual UGT-cDNAs in bakers yeast to phenotypically test and validate the resistance properties.
