Liposomes for biopharmaceutical application prepared by oriented sonication
Abstract
For liposome preparation only few techniques are available. Most of these methods are based on technologies were lipids are dissolved in organic solvent or detergents prior liposome formulation. All the used solvents needs to be removed quantitatively when the final product is used for therapeutic application. Alternative techniques are based on sonication and/or cavitation which may induce product degradation, mainly generated by oxidation processes. The aim this project is the preparation of liposomes with controlled, homogenous ultrasonication, which is generated by an ultrasonic control unit. Therefore, lipids were suspended in an aqueous phase and by controlled sonication, bilayer membranes are formed. Lipid, protein and bilayer characteristics will be analysed to study product properties in dependence on varying physical parameters. As a model protein, the recombinant, human Cu/Zn Superoxide Dismutase is used.
