Abstract
In absolute protein quantification, there is a demand for introducing reference methods and standards, which can be generalised to a wide range of proteins. Traceability of values assigned to calibrants, following the principles of metrology needs to be established. We propose the integration of LC-ICP-MS based sulphur speciation analysis, as generic absolute protein quantification method. This concept enables isotope dilution strategies, the quantification method of choice when aiming at accuracy. Species specific standards will be produced for methionine and cysteine by in vivo synthesis using 34S as sulphur source in yeast fermentations.
